Nucleus of Allium cepa, Viewed with a UV-Microscope

a: Wave length lambda = 310 nm, b: Wave length lambda = 280 nm. The contrast increases with decreasing wave length. The nucleic acids have an absorption maximum of 260 nm. Still, it is recommendable to work with UV radiation of rather long waves since the decrease of wave length is coupled to an increase in cell damage. (I. K. LICHTSCHEIDL, 1987)
