The UGC is developing an integrated random and directed sequencing strategy for high
throughput sub-cloning, mapping and sequencing of both BAC clones and microbial
genomes. Key features are:
- Efficient generation of 10-12 kb sub-clones on low-copy/copy-inducible plasmids
- High throughput, multiplex library insert sizing, transposon bombing and mapping
- Automated clone storage/retrieval and DNA preparation for sequencing
- Integrated software tools for clone tracking, process control and sequence assembly/finishing
