IN VITRO CONSERVATION OF GENTIANA LUTEA L.

Extracts of roots of Gentiana lutea are widely used in the traditional medicine in Ukraine as a tonic, eupeptic and digestive stimulants as well as anti-jaundice, antiseptic, antiphlogistic, antimicrobial substances. There is an increasing interest in the constituents of gentians as these plants are rich in resources of xanthones, potential antidepressant drugs. G. lutea have been established as a excised root-organ culture on the liquid 1/2 Murashige and Skoog (MS) medium without phytogormones. After 6 weeks of subcultivation on this medium, fresh weight of excised roots had increased up to 3g. MS medium with 1/2 content of salts and double concentration of CaCl2 and supplemented with BAP 0,05 mg/l, kinetin 0,1 mg/l had proven to be optimal for shoot multiplication. Rooting of G. lutea plantlets was achieved on 1/2 strength of MS medium containing 0.1-0.3 mg/l NAA. The maintenance of callus cultures (root and cutting explants) was performed on the 1/2 MS medium supplemented with BAP 0.2 mg/l and 2,4-D 0.1-0.9 mg/l.
