RAPD ANALYSIS IN SUBTRIB MICHELIINAE

Zhu Jianming, Wang Ting, and Su Yingjuan, School of Life Sciences, Zhongshan University, Guangzhou 510275, P. R. China

The total DNA were isolated by CTAB method from fresh leaves of subtrib Micheliinae including 14 species from 3 genera as amlification templates and 13 primers selected from 3 Operon RAPD kits(A,C and E)could amplify good RAPD products. The dendrogram constructed from genetic distance through UPGMA method shows that subtrib Micheliinae can be divided into two genera. One is subgenus M.figo including M. martinii, M.chapensis, M. yunnanensis and M. figo, the other is subgenus M. alba which is divided into two section-Section M. floribunde including M. floribunda, M.balansae, M. macclurei, M. crassipes and T.odorum, while Section M.alba including M. alba, M. champaca, M. maudiae and M. foveolata. Based on the RAPD analyses, T.odorum is not supported to be separated from genus Michelia and considered to be a genus which is discordant with Liu Yu-hu*s system. However, it is supported to treat P.baillonii as a single genus independent of genus Michelia which is appropriate to Liu*system.Comparing these results with Nooteboom*s or Dandy*s system, some are accord with the two systems while others are not.
