ORGANOGENESIS AND PLANTLET FORMATION IN VITRO IN CHUKRASIA TUBULARIS ADR. JUSS.

T. Pullaiah and M.A.H. Nagalakshmi (Department of Botany, Sri Krishnadevaraya University, Anantapur, 515 003, India)

Adventitious shoots were initiated from cotyledon explants of seedlings of Chukrasia tabularis, when cultured on MS medium, the best suitable medium, supplemented with BAP (2 mg/1), KN (1 mg/1) and IAA (0.5 mg/1) after four weeks. However, shoot bud formation was accompanied by intervening callus. Sucrose was found to be the best source of carbon followed by dextrose and fructose and also the percentage of sucrose made a marked difference in the nature of shoots formed. Among the three concentrations tested, i.e. 2%, 3% and 4%, only at 4% healthy shoots were formed. Most of the shoots were of 2 cm long and these were transferred to rooting medium consisting of MS basal medium with IAA (3 mg/1). After the onset of rooting from the basal end, these were transferred to agar solidified soil solution (10% soil), where elongation of roots occurred. Experiments with reference to survival efficiency of these plantlets are to be carried out.
