MAP-BASED CLONING OF THE BARLEY STEM RUST RESISTANCE GENES RPG1 AND RPG4.

High resolution genetic and physical maps for the barley stem rust resistance genes Rpg1 and rpg4 were developed. Map-based cloning was facilitated by the resources of the small genome cereal rice and a 6.3X barley BAC library. A rice BAC containing the closest Rpg1 flanking markers was sequenced. Analysis of the 33 kb region revealed 5 genes, but a rice Rpg1 homologue was not identified. Four of the five genes showed a high degree of similarity with genes on Arabidopsis chromosome 4. We have identified RFLP markers bracketing the rpg4 locus that map about 0.5 cM apart and established rice and barley BAC contigs across this region. The preliminary data indicate that the flanking markers are less than 100 kb apart in rice and less than 200 kb apart in barley.
