STUDIES ON ARTIFICIAL SEEDS OF DENDROBIUM CANDIDUM WALL.EX LINDL.

Dendrobium candidum artificial seed was studied based on the researches of the D.candidum seeds germination,suspension-cultured protocum-like bodies(PLBs),seed coat material and encapsulating method. 2.4-D stimulated germination only in 0.5ppm concentration,within the range from 0.5ppm to 2ppm,NAA raised the seed germination rate,ABA was important to synchronize the PLBs and to improve their qualities. The multiplication of PLBs were affected significantly by the culture conditions.Compared with the PLBs multiplicatiing speed in stationary liquid medium,agitated liquid medium and solid medium, we found the agitated liquid medium was best,and the optimal was MS(1/4NH4CO3) with 400ppm Vc which could effectively prevent PLBs from turning brown during the suspension culture.The germination rate of artificial seeds made with 3% sodium alginate and 2% calcium chloride reached 85%,and the germination rate of the two kind of artificial seeds encapsulated with clay and vermiculite were almost same. The desiccation of PLBs,selection of preservative,and the affect of chitosan as the outer seed coat to the artificial seed germination were discussed.
