IN VITRO CULTURE AND CALLUS INDUCTION OF MARRUBIUM VULGARE AND POLYGALA CYPARISSIAS

Pharmacological analysis of Marrubium vulgare L. (Horehound) have demonstrated antispasmodic effects, expectorant and analgesic. Hydroalcoholic extracts of Polygala cyparissias exhibits potent antinociceptive and antiedematogenic activities. The aim of the present study was to initiate an efficient method for in vitro culture and callus induction in this species. Seeds were surface sterilezed and germinated in hormone free MS. The plants were kept in a culture room at 25?C and photoperiod of 16hs. Callus induction were initiated from the medium supplemented with 0.25, 0.5 and 1.0 mg.l-1 2,4-D. The methodology used for initiated in vitro culture of M. vulgare and P. cyparissias was efficient, and provided development of 100% of explants. The 2,4-D auxin was efficient in callus induction at all concentration, and provided significant callus fresh weight development of them, emphasized the concentration of 0.25 mg.l-1 at both species.
