Variation in nuclear 2C DNA content of seven amaranth species was explored for resolving phylogenetic relationships. Measured by flow cytometry, DNA content of three grain species and Amaranthus hybridus was nearly identical (1.26-1.27pg), but the value was lower in A. quitensis (1.19pg) and A. powellii (1.22pg) and much higher in A. tricolor (1.81pg), supporting that A. hybridus is the common ancestor of grain amaranths. Based on DNA content, copy number of two copia-type retrotransposons was estimated, and the utility of partial retrotransposon sequences was demonstrated as probes for fingerprinting analysis of amaranths.
