XVI International Botanical Congess
Various explants of C. alata and A. amara were evaluated for their morphogenic potential under in vitro culture conditions. Axillary buds and shoot tips were collected from aseptically raised seedlings and grown in MS medium supplemented with various concentrations and combinations of growth regulators for shoot multiplication and rooting. Multiple shoots of both the plants were induced in the cultures of axillary buds and shoot tips on MS medium containing BA 1.0, 1.5 and IAA 0.2 and NAA 0.2 mg/l respectively. The shoots of C. alata and A. Amara were rooted on 1/2 strength MS medium containing IBA 0.5 and 0.8 mg/l respectively. The regenerated plantlets were later acclimatized in the greenhouse. Antioxidants were used to prevent browning and favored organogenesis. Anatomical studies revealed that meristomoids located at the sub epidermal level were differentiated into shoots. Isozymes were also studied to test the clonal purity.
