Ps1-barnase and phcintG genes were co-transferred into rice genomic DNA and male sterile (MS) rice plants were induced. Primary callus and suspension cells as the target tissues were bombarded with the plasmid DNA mixed to Gold Particle. Among the transgenic plants positive for ps1 gene, we found complete-, high- and semi-MS plant. The complete MS plant produced no-function pollens and could not set seed in the condition of self-pollination. While the plants negative for barnase gene expressed fertile showing that the MS of the transgenic plant was caused by ps1 gene and the gene expressed well in rice. Southern blot analysis from genome DNA of the plants in T1 generation showed that 3/4 plants were positive and 1/4, negative. The segregation-ration of fertility expression in T1 and back-cross F1 generations was 3 sterile: 1 fertile and 1 fertile: 1 sterile respectively. These results showed that the foreign gene ps1-barnase was integrated in rice genome well and transferred to the offsprings as a single site stable.
