SELECTION AND CHARACTERIZATION OF SALT RESISTANT VARIANTS AND PLANTREGENERATION OF WILD BARLEY

Mature embryos of wild barley (Hordeum brevisubulatum) were used as explants to induce calli, embryogenic suspension cell cultures were established. By treating suspension cells with ethylemethane sulfonate, NaCl tolerant variants were selected from the saline medium. The results show that the variants could grow on 1.2% NaCl medium. Physiological and biochemical analysis showed that free L-proline in 1.2% NaCl tolerant calli was as much as 6.68 times of the control, and relative conductive ratio was 29.92% lower than the control. Its esterase isozymes were different from the control, with two extra isozymes bands. All analyses showed that NaCl selected calli have the characters of NaCl-tolerance. Regenerated plants were different from NaCl tolerant calli.
