REGENERATION OF HELICONIA BURLEANA ABALO & MORALES BY MERISTEM AND SEXUAL SEED IN VITRO CULTURE

M.E. Corrales (Laboratorio de Cultivo de Tejidos Vegetales, Departamento de Biologia, Universidad de Antioquia, A.A. 1226, Medellin-Colombia, Fax: 574-2638282, e-mail: cmonica@lettera.net). Key words: Heliconia burleana, micropropagation, in vitro tissue culture, plaianillos

Apical and auxiliary meristems were cultured in MS medium supplement with myoinositol 0.1 g/L, glutamic acid 0.05 g/L, adenine 0.005 g/L, sucrose 30 g/L, agar 6.0 g/L and growth regulators (BAP and AIA) in different concentrations. Statistical analysis was based in t assimilated" Test. The two treatments with BAP 4.0 mg/L, AIA 1.0 mg/L and BAP 6.0 mg/L, AIA 1.5 mg/L showed the best shoots production, specially the auxiliary meristems that gave rise to a better response. In vitro seedling are still in production with vigorous appearance and dark green color. Additionally, in vitro seed culture was carry out using several mechanical scarification and aseptic treatment with hipocloride at 2% and then seed were place in MS medium without any growth regulator. Germination took place between 15 to 45 days after cultured and 40% of germination was achieved with this medium. Finally, seedlings were transferred to the new MS medium supplemented with BAP 3 mg/L. After two months the plantlets give rise to multiple shoots.
