Arabidopsis ß-thioglucoside glucohydrolase1 (TGG1, or myrosinase) localization was studied by using TGG1 promoter-gus gene fusion system. The guard cell specificity of gus gene expression was found in all guard cell containing organs, such as cotyledons, leaves, and inflorescence stems of all transgenic Arabidopsis lines, containing TGG1 promoter-gus gene cassette. The guard cell specific expression of TGG1 gene in Arabidopsis was confirmed by the in situ hybridization experiment using TGG1 gene specific probe. Transgenic tobacco plants, containing the same DNA cassette, also showed the guard cell specific expression of GUS enzyme. This suggested that TGG1 promoter could be expressed in some other plants that do not have myrosinase-glucosinolate system, and conserve the guard cell specific expression property.
