CLONING OF A NOVEL PROTEIN WHICH BINDS TO THE ENHANCER OF CHLOROPLAST psbD LRP

The expression of the chloroplast psbD gene which encodes the D2 protein of photosystem II is mainly regulated by a phylogenetically conserved light responsive promoter (LRP) in mature chloroplasts. Transcription from LRP is strongly enhanced by an upstream sequence which binds an unidentified DNA binding protein. In this study, we obtained an Arabidopsis cDNA encoding a protein that binds to this sequence by one-hybrid system. It contains a putative DNA binding domain which has high homology to several DNA binding proteins. mRNA of this protein was mainly accumulated in leaves. Incorporation of this protein fused with GFP into chloroplasts was observed by transient assay. Phenotype of an arabidopsis mutant lacking this protein and sequence-specific DNA binding activity of this protein is further examined.
