GENERATION OF NUCLEAR MALE STERILITY IN RICE (ORYZAE SATIVA)

The aim of this study is to generate nuclar male sterility for hybrid seed production in commercial varieties of rice. For this, we have isolated a cDNA clone,Ory s 1, from rice anthers cDNA libraries. The isolated cDNA encodes for a protein of approximately 35kD and results from RNA blot analysis showed that this gene is only expressed in mature anthers. To study the activity of Ory s 1 promoter during pollen development, two types of constructs, Ory s 1- GUS and Ory s 1 - GFP were made and tested using transient expression assays. The results of these experiments showed that Orys 1 promoter is pollen specific. In order to study the role of Ory s 1 in pollen development, antisense Ory s 1 construct was introduced into rice via Agrobacterium-mediated transformation. Rice plants regenerated from these experiments are being grown under glasshouse conditions. Pollen from these transgenic plants will be analyzed for pollen fertility.
