Pharmacological analysis of M. vulgare L. (Horehound) have demonstrated antispasmodic, expectorant and analgesic effects. Hydroalcoholic extracts of P. cyparissias exhibits potent antinociceptive activities. The aim was to initiate an efficient method for 'in vitro' culture and callus induction in these species. Seeds were germinated in free MS medium hormone. The plants were kept at 25°C and photoperiod of 16hs. Callus induction were initiated from the medium supplemented with 0.25, 0.5 and 1.0 mg.l-1 2,4-D. The methodology used for initiated 'in vitro' culture of M. vulgare e P. cyparissias was efficient, and provided development of 100% explants. The 2,4-D auxin was efficient in callus induction at all concentration, and provided significant callus fresh weight development, emphasizing concentration 0.25mg.l-1 on both species.
