A PAC CLONE, PMM4G7, IS DERIVED FROM Y CHROMOSOME OF MARCHANTIA POLYMORPHA AND CONTAINS MALE-SPECIFIC REPEATED SEQUENCES AND A PUTATIVE GENE CODING FOR A RING-FINGER PROTEIN

M. polymorpha is dioecious, and its sex is determined by sex chromosomes. To isolate genes responsible for sex determination and sexual differentiation, genomic libraries of male and female M. polymorpha were constructed. A clone, pMM4G7, was isolated from the male library by differential screening using male or female total DNAs as probes. The labeled pMM4G7 hybridized specifically to the male total DNA. pMM4G7 was confirmed to be localized on the Y chromosome by FISH. Analyses on pMM4G7 subclones revealed the presence of novel repeated sequences and an open reading frame containing a RING finger motif in the Y chromosome.
