STRUCTURAL ASPECTS INVOLVED IN BANANA BUDS FORMATION IN VITRO

Jane Elizabeth Kraus*, Gilmar Zaffari, Gilberto Barbante Kerbauy, Elaine C. Romano (*Instituto de Biociências, USP, São Paulo, Brazil)

Shoot apexes of Musa acuminata (AAA) cv. Grande Naine were used for explant. The explants were cultivated in MS medium (1 mg.L-1 BAP) during 60-day culture and transfered to MS (2.5 mg.L-1 BAP). Samples were taken after 0, 30, 60, 65, 70 and 75 days of culture Structural analysis of explants showed alterations on size and shape of the apical meristem of shoot. The explants had undergone a radial growth, proportionally to the period of culture (0-60days). Between 65-75 days of culture two kinds of adventitious buds were formed: buds formed at the base of leaf sheath (lateral buds) of the explants and buds formed on tissue proliferated from the explant. Binucleated cells were noticed on the meristem of the bud. The buds formation and their implication in somaclonal variation are discussed (CNPq, FAPESP).
