Sequences of matK gene and rDNA ITS regions from the family Cercidiphyllaceae, the genus Daphniphyllum (Daphniphyllaceae) and all six subfamilies of Hamamelidaceae were determined to study the phylogenetic relationships among those taxa. Maximum likelihood and neighbor-joining trees were constructed based on ITS and matK sequences respectively. Similar topologies were obtained. Cercidiphyllum and Daphniphyllum were closely related to the family Hamamelidaceae (excl. Liquidambaroideae) with high bootstrap values. Cercidiphyllum was relatively closed to Mytilarioideae. The topologies showed there are close relationships among Disanthoideae, Hamamelioideae, Mytilaroideae, Exbucklandia, Rhodoleia, Daphniphyllum, Cercidiphyllum, and Liquidambaroideae was distantly related to them.
