XVI International Botanical Congess
As current research focus at the IPK Genebank, the genus Lactuca (830 accessions) is analyzed by PCR-based methods. At first, 83 name duplicates from 34 varieties are examined to confirm their supposed identity. Although entries within most varieties are identical or very similar, 1/3 of the varieties show differences. This recommends analyses of every name duplicate before deciding to discard all but one. In perpetual kale, RAPD analyses yield only 7 different patterns in 64 genebank accessions. Compiled with additional data from CPRO-DLO (NL), up to 22 entries display identical features, offering possibilities to reduce the number of propagated clones. Finally, Solanum nigrum complex species are analyzed for taxonomic and characterization aims. By applying 2 SSR primer pairs on 18 entries from 4 species, all species can be differentiated. On the intraspecific level, AFLPs seem preferable to RAPDs.
