Subcellular localization of phosphoglycerate kinase (PGK) in Chlamydomonas reinhardtii was studied by immuno-electron microscopy. When antibodies raised against chloroplastic phosphoglycerate kinase (chl-PGK) was used, signals were found only in the chloroplasts especially concentrated in stroma. No signal was detected in the cytoplasm. In Southern blot analysis using chl-PGK cDNA as a probe, only one band (4.2 kb Bam HI fragment) was detected. So far only one form of PGK was observed in Chlamydomonas reinhardtii and analysis of 4.2 kb fragment is underway.
