VARIATION IN THE ITS OF CULTURABLE TRICHOMYCETES

Trichomycetes consists of three orders, of which only Harpellales contains culturable species. More than 70 isolates, distributed worldwide, representing ca. 30 described species from 4 genera (Furculomyces, Smittium, Simuliomyces and Genistelloides) were studied. Their internal transcribed spacer regions (ITS 1 and ITS 2) were analyzed by PCR amplification and RFLPs. The aim was to assess the variation of the ITS regions to determine their suitability in phylogenetic studies. Each PCR product was digested with several restriction enzymes and the restriction patterns were compared by UPGMA. The analyses revealed high degree of polymorphism among Smittium species (e.g.: the ITS 1 length ranges from 450 to 950 bp and the ITS 2 from 650 to 1000 bp). They also allowed separation of isolates into distinct groups, that in general correlated well with morphologically defined species.
