A PHYLOGENETIC STUDY OF GEOGLOSSUM AND TRICHOGLOSSUM BASED ON ITS AND GROUP I INTRON SEQUENCES

Geoglossum and Trichoglossum (Helotiales) are closely related genera based on morphological characters. In this study, PCR amplifications using primers ITS4 and ITS5 yield two fragments in Geoglossum and Trichoglossum. One fragment is around 500-600bp, which is similar to that obtained from the other members of the order, the other is around 500bp longer. Sequencing the longer fragments revealed that Group I introns are inserted 31bp from the 3' end of the SSU rDNA, which is position 1506 corresponding to E. coli SSU rDNA. Both the ITS and intron trees suggest that species of Geoglossum and Trichoglossum form a monophyletic group, but within this group some Trichoglossum species are closer to certain Geoglossum species than to other species of Trichoglossum. The P-TPT Test suggests that the ITS and intron data are combinable giving better support.
