Cross-sections of the East African Apocynaceae Tabernaemontana pachysiphon show that the cuticle of the leaf undersurface covers the foldings of the epidermal cell walls like a tablecloth. It is stained with Sudan IV, a classic maker.
The fluorescent dye acridine orange is exclusively used for the staining of the cuticle.
The epidermal cells of the upper leaf surface are also structured. The cuticle is so thick that it levels out the unevenness of the cell walls. The walls are stained with methylene blue.
